Increasing the pH influenced the physical character-
istics of the granular sludge.The TS content decreased
by 50% from 114.5 g TS 1-1 granules to 55.1 g TS 1-1
granules during a stepwise pH increase from 7.3 to 8.2.
Likewise,the VS content decreased from 39.2 g VS 1-1
granules to 11.2 g VS 1-1 granules,while the ash con-
tent increased from 65.8% to 79.6% TS.
The pH increase resulted in formation of a higher
fraction of granules with a smaller diameter and a si-
multaneous decrease in the fraction of the larger gran-
ules (Fig.4).The smallest particles,with a diameter less
than 0.4 mm,constituted 53% of the granules at pH 7.3.
This group of granules increased to more than 60% at
pH 7.9 and pH 8.2.The fraction of the granules with a
diameter between 0.4 and 0.8 mm also decreased when
the pH was increased.The density of granules de-
creased from 1067 g 1-1 at pH 7.3 to 1017 g 1-1 at pH
8
For 2 months (pH 7.3-7.9) the organic loading rate (LR) was
maintained at 16 g COD 1-1 day and 89% of the effluent was re-
circulated.Months of effective performance (pH 7.3 to 7.9) was
followed by an accident after 4 days of operation at pH 7.9 due to
a breakdown of the influent pump system.During the accident
approx.35% of the biomass was washed out of the reactor.The
performance of the reactor at pH 7.9 was re-established after 2
months operation at an LR of 10 g COD 1-1 day and the loading
rate was kept at this value for the rest of the experiment.
Medium.The general composition of the fish condensate is shown
in Table 1.The condensate was supplemented with
K2HPO4.H20,2.0mM; MgC12,0.49 raM; CaC12,0.75 mM; KC1,
3.6 mM; FeSO4.H20,0.020 mM; and H2SO4,0.015 mM.Trace me-
tals solution was added as described by Angelidaki et al.(1990).
Batch tests.The specific methanogenic activity (SMA) of the gran-
ular biomass was tested in batch experiments at different pH val-
ues from 7.1 to 8.5.The experiments were performed at the begin-
ning of the pH acclimatization period and repeated at the end of
the UASB reactor experiments when the reactor pH was 8.2.
The batch experiments were performed in 58-ml vials contain-
ing 20 ml fish condensate supplemented as described above.A
phosphate buffer system (18 mM) was used and the pH was
checked before and after the experiments; the pH was constant
throughout the experiments.The vials were incubated at 34°C
and all experiments were performed by measuring initial methane
production in triplicate.
